An article by Cornerstone explains the possibilities of CBD soothing the stomach and controlling intestinal inflammation. More than 200,000 times each year in the United States, people are diagnosed with Irritable Bowel Syndrome (IBS). Unfortunately, IBS is not well understood, and no cure currently exists. However, some treatments have been shown to ease the symptoms.

The article notes that several studies have shown that CBD helps with healing inflammation and restoring normal intestinal motility, but it’s almost all done in rodents – not people. Studies are currently being conducted to establish whether the results are the same in human subjects.

A research group from Naples, Italy set out to test the theory that CBD extract would have the same result. A full spectrum product was used in this research, meaning not just CBD was used, but all of the cannabinoids within the cannabis plant. These variety of cannabinoids are believed to create an “entourage effect”. Various tests have shown that, especially for pain and inflammation, that this entourage effect had more of an impact than using CBD alone, without the combination of others.

Mice were administered colitis or intestinal inflammation via administration of DNBS (dinitrobenzenesulfonic acid). DNBS is an inflammatory chemical that has been shown to damage the intestines, but rarely leads to fatality. They used a decrease in body weight and increase in colon weight/length to measure inflammation. CBD was found to not modify the loss of body weight, but reduced the inflammation of the colon. Interestingly, pure CBD had no effect on either parameter – only the full spectrum products had an impact.

When measuring intestinal transit, the goal was to meet the ideal criteria for an IBS medication – ease pain and restore motility. Researchers fed mice food mixed with charcoal, and then at the end of the study, after the mice had deceased, would measure how far the leading edge of the charcoal had moved, dividing this distance by the total length of the colon in each mouse. “To mimic the accelerated intestinal movement caused by IBS, researchers applied croton oil, a particularly pungent oil from the croton tree, then applied CBD and CBD extract.” CBD extract, as before, reduced the excess movement brought on by croton oil. Pure CBD did duplicate this effect.

The conclusion of this article is that oral administration of CBD and CBD extract (full spectrum) can be effective as body cavity injection in rodents – and would suggest the same in humans. There was a large performance difference between pure CBD and the more effective full spectrum CBD oil.

Cannabinoids and the Endocannabinoid System – Tumors and Cancer

We probably all know someone that has suffered from cancer at some point in our life. It is a ferocious disease that often has devastating impacts on individuals, families, and even societies. Cancer claims nearly 40% of it’s victims, according to the National Cancer Institute. Additionally, it is expected that 39.6% of men and women will develop some form of cancer during their lifetime.

So, can cannabinoids, and specifically cannabidiol (CBD), play a role in minimizing these numbers? This 2006 study found that of the five cannabinoids tested, cannabidiol (CBD) was the most potent inhibitor of cancer cell growth. Additionally, the study found the following to be true:

“At least for MDA-MB-231 cells, however, our experiments indicate that cannabidiol effect is due to its capability of inducing apoptosis via: direct or indirect activation of cannabinoid CB(2) and vanilloid transient receptor potential vanilloid type-1 receptors and cannabinoid/vanilloid receptor-independent elevation of intracellular Ca(2+) and reactive oxygen species. Our data support the further testing of cannabidiol and cannabidiol-rich extracts for the potential treatment of cancer.”

While the study above was more focused on breast cancer, there are studies like this one from 2011 that show similar results for those with prostate cancer. This study concluded that:

Cannabinoid receptor agonists induce phosphatases and phosphatase-dependent apoptosis in cancer cell lines; however, the role of the CB receptor in mediating this response is ligand-dependent.

Both of these studies reference apoptotic occurrences – a process that marks cells for scheduled death. It is sometimes said that these cells then “commit suicide” after being marked. Clearly, there is evidence showing that cannabinoids, and specifically CBD can be helpful to those with cancer.

Lastly, another study showed how CBD and other cannabinoids may be helpful in treating cancer pain. In this double-blind study, CBD was shown to reduce the Numerical Rating Scale (NRS) by more than 30% when compared to the placebo. In conclusion the study states:

This study shows that THC:CBD extract is efficacious for relief of pain in patients with advanced cancer pain not fully relieved by strong opioids.

It was able to inhibit not only the actions of endocannabinoids but also the effects of other inflammatory mediators such as steroid hormones that stimulate fat production in these cells. “Another important result: we found that low concentrations of CBD were very effective in inhibiting lipid synthesis but did not affect the viability of cells. This was also in contrast to the effects of Vitamin A derivatives which inhibit the fat production of sebocytes by killing them.”

The study then goes on to explain that in the right concentration, CBD does not only inhibit the bad cells, but leaves the good cells alone. The article continues by asking this question:

Cannabinoids Impact on Celiac Disease

Celiac disease has recently become a well-studied idiopathic disorder. According to the Celiac Disease Foundation, 1 in every 100 people is at risk for celiac disease.  Worse, first degree relatives of someone with celiac have a 1 in 10 chance to develop the disease. Around 80% of people with celiac disease don’t know they have it and may be asymptomatic or underdiagnosed.

So, how can cannabinoids impact those struggling with the disease? One study shows that those with celiac disease, and are untreated, have a much higher level or cannabinoid receptors CB1 and CB2. As this study points out:

“Our data show higher levels of both CB1 and CB2 receptors during active disease and normal CBR levels in treated celiac patients. In conclusion, we demonstrate an up-regulation of CB1 and CB2 mRNA and protein expression, that points to the therapeutic potential of targeting CBR in patients with celiac disease.”

In short, there are more CB1 and CB2 receptors available in those with untreated celiac disease. This means that cannabinoids, such as cannabidiol (CBD) will be received more efficiently.

Additionally, it can be implied that someone who eats gluten accidentally would then have an increase in these receptors as well. Cannabinoids For Neuropathy

Over 16 million people in America suffer from neuropathic pain. This pain stems from either nerve injury, disease states, or toxic insults. A study from the National Institutes of Health researched whether cannabinoids, like CBD, could be used to address this unmet need. The results are pretty remarkable.

In this double-blind, randomized, placebo-controlled, parallel group study, it was analyzed whether cannabinoids could help peripheral neuropathic pain (PNP) treatment.  For 15 weeks, the group study was analyzed and investigated. Over 300 people with peripheral neuropathic pain. This pain was associated with allodynia because it poses a significant clinical challenge.

Of these 303 people, 128 were randomized to cannabinoids, and 118 to placebo, in addition to their current analgesic therapy. Not only was there a statistically significant treatment difference favoring cannabinoids, but there was also a reduction in mean PNP scores in both treatment groups. However, the reduction was greater in the ones taking cannabinoids. Additionally, sleep quality was measured and improved greatly in favor of the cannabinoid treatment.

The study concluded by saying that these findings demonstrate that, “in a meaningful proportion of otherwise treatment-resistant patients,” there was improvement in pain, sleep quality, and Subject Global Impression of Change (SGIC). It also noted that the cannabinoids were well tolerated with no new safety concerns identified.

So, although this was only a study of 300 people. It did appear that cannabinoids were effective in treating, or at least diminishing the neuropathic pain experienced by these people.

A different study, examined whether cannabinoids would suppress chronic inflammation and neuropathic pain. In short, again, it found that they do. It states that:

“The cannabinoids significantly potentiate glycine currents in dorsal horn neurons in rat spinal cord slices. The analgesic potency of 11 structurally similar cannabinoids is positively correlated with cannabinoid potentiation of the α3 GlyRs. In contrast, the cannabinoid analgesia is neither correlated with their binding affinity for CB1 and CB2 receptors. NMR analysis reveals a direct interaction between CBD and S296 in the third transmembrane domain of purified α3 GlyR. The cannabinoid-induced analgesic effect is absent in mice lacking the α3 GlyRs. Our findings suggest that the α3 GlyRs mediate glycinergic cannabinoid-induced suppression of chronic pain. These cannabinoids may represent a novel class of therapeutic agents for the treatment of chronic pain and other diseases involving GlyR dysfunction.”

It’s that time of the year. Whether we like it or not, it’s “flu season”. Coughing, fatigue, aches, pains, nausea and vomiting are all miserable – and flu season has been making national news almost daily. So, is there actual hope that CBD may help fight the flu?

One of the ways that CBD may help reduce inflammation is by preventing mast cells from releasing histamine into the blood stream. Because histamines are the chemicals which spur an inflammatory response, this can play a vital role in reducing inflammation.

This article describes an account of using CBD for this exact reason. CB1 receptors are found in our brain, according to a study by the Journal of Allergy and Immunology referenced in the article. CBD1 receptors may be useful in managing allergies and skin diseases (mainly rashes and eczema). The stimulation and inhibition of CB1 receptors were directly linked to mast cell activity.

Another study referenced by this article is by the Journal of Pharmacology and Therapeutics found that CBD reduced airway obstruction caused by raw egg whites. Ovalbumin, uncooked egg white, is a known mast cell trigger and histamine liberator. It is used in studies to cause inflammation and histamine release, with the hopes of finding how to reverse it. The Journal of Biochemistry found that cannabinoid stimulation of the CB1 receptor prevented mast cell degranulation – the process which releases histamine inflammation into the blood stream.

So, as with most research regarding CBD, hemp, and cannabis, this research and studies are anecdotal, and have not been tested on a large scale. The Journal of Endocrinology tells us that in diseases like asthma, arthritis, inflammatory bowel disorders (IBS), there is regular, unnecessary activation of mast cells.

The Journal of Bone and Mineral Research published a study regarding CBD and it’s ability to enhance fracture healing and stimulate lysyl hydrozylase activity in osteoblasts. Cannabinoid ligands help regulate bone mass. With over 6.3 million bone fractures a year, the following research is very promising for those suffering from one. Although they all vary in severity and placement, one thing they almost all share is immobilization and discomfort.

One of the most more serious bone fractures are femoral fractures. This study from the Journal of Bone and Mineral Research reports that CBD enhances certain biomechanical properties of healing rat mid-femoral fractures. CBD was shown to increase the maximal load and work-to-failure, but not the stiffness of femurs. Tetrahydrocannabinol (THC) did not share this effect.

Using micro-computed tomography, CBD was shown to transiently reduce the fracture callus size after just four weeks, but took 6-8 weeks to reach control level. The density of the callus material was not effected.

Lysine hydroxylation is involved in collagen crosslinking and stabilization. So, what does CBD have to do with lysine hydroxylation? “CBD stimulated mRNA expression of Plod1 in primary osteoblast cultures, encoding an enzyme that catalyzes lysine hydroxylation”. CBD was shown to also increase the collagen crosslink ratio, which is believed to contribute to the improved biomechanical properties of the fracture callus.

The study concluded by stating: “Taken together, these data show that CBD leads to improvement in fracture healing and demonstrate the critical mechanical role of collagen crosslinking enzymes.”

Although there is more research needed to conclusively state whether CBD helps with these issues, this anecdotal research is a great step in the right direction.
